Amos Kirkpatrick was born in Camden District, South Carolina on 30 May 1779. Amos appears on the tax list of Green County, Kentucky in 1800. By 1803 he is found in Jackson County, Tennessee, where he lived the rest of his life. He died 1 or 9 December 1861, at Big Bottom, in Jackson County. His profession is unknown.

Amos fought in the War of 1812 and was with Andrew Jackson at New Orleans. He was 1st Sergeant in the Company of Captain Henry West. A list of members of that company was copied at one time from the diary kept by Amos Kirkpatrick, but the diary itself has not been found. This company met at the mouth of Roaring River, 13 November 1814, and arrived at Nashville on 21 November. They were mustered into service at Nashville on November 23. They left for New Orleans November 24 and took part in the Battle of New Orleans on 7 and 8 January 1815.

Amos' will is dated 7 June 1856. After his death, some of his heirs contested the will. In papers filed in the Jackson County, Tennessee court, it was asserted that : "Said paper purporting to be the will of said Amos Kirkpatrick, deceased. . . is not the will of said Amos Kirkpatrick, deceased. They, your petitioners state to your worships that all the time said paper writing bears date, dates for some time previous thereto and after said date up to the time of his death said Amos Kirkpatrick was not of sound mind and disposing memory. He was so far demented and imbecile as to be incapable of comprehending his property or the objects of his bounty. He was wholey incapable of transacting his business affairs guarding his interest or taking care of or making a suitable and just dispostion of his property by will ortherwise on account of his mental inbecility. . ." The conclusion of this lawsuit is not clear, but from two letters written by his son Nero to Amos, one in 1856 and one in 1857, it would appear that this son, at any rate, felt his father to be competent to discuss business dealings.

Amos married three times. His first wife was Sarah Green Ellis, daughter of Ephraim Ellis, who was born 10 May 1779, Camden County, South Carolina, although one souce says she was born in Rutherford County, North Carolina. She died in 1823. They were married 27 November 1798 in Green County, Kentucky.
